Digital audio cannot be processed sample-by-sample; modern CPU operating systems process audio in discrete chunk packages called buffers. The fundamental time required to fill a single audio buffer is calculated as:
Frame Duration (ms) = (Buffer Size in Samples / Sample Rate in Hz) × 1000
For instance, at a standard studio sample rate of 48,000 Hz with a buffer size of 128 samples: (128 / 48000) × 1000 = 2.67 milliseconds.

If a 128-sample buffer is only 2.67ms, why do smartphone voice changers measure 35ms–45ms overall? Because the complete roundtrip chain includes 5 hardware and software stages:
